Substrate specificity of fatty acid synthetase from yeast.

چکیده
The purified multienzyme complex fatty acid synthetase from yeast synthesizes palmitoyland stearoyl-CoA with NADPH, ace@-CoA and malonylCoA as substrates. At least seven different enzymatic steps have been shown to occur in the overall synthesis by the use of appropriate model substrates [l] , .e.g. S-acetoacetyl-N-acetylcysteamine for the @-ketoreductase partial activity. As reported in this communication, the model compounds S-acetyl-N-acetylcysteamine and S-m~onyl-~-acyIcysteam~e can replace the natural substrates acetyl-CoA and malonyl-CoA in the overall fatty acid synthesis.

Fatty acid structural requirements for activity of arachidonoyl-CoA synthetase.
We have examined the fatty acid substrate specificity of arachidonoyl-CoA synthetase from human platelet membranes. A variety of positional isomers and chain-length analogs of arachidonic acid [20:4(5, 8, 11, 14)] were synthesized, and assayed for their ability to inhibit arachidonoyl-CoA formation or to serve as substrates for the synthetase. The chain-length specificity of the synthetase for ...
متن کاملArabidopsis contains nine long-chain acyl-coenzyme a synthetase genes that participate in fatty acid and glycerolipid metabolism.
Long-chain acyl-coenzyme A (CoA) synthetases (LACSs) activate free fatty acids to acyl-CoA thioesters and as such play critical roles in fatty acid metabolism. This important class of enzymes factors prominently in several fatty acid-derived metabolic pathways, including phospholipid, triacylglycerol, and jasmonate biosynthesis and fatty acid beta-oxidation. In an effort to better understand th...
متن کاملMetabolism of N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide by prostaglandin endoperoxide synthetase.
Cooxidative metabolism of the urinary bladder carcinogen N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide (FANFT) was examined using solubilized and particulate microsomal preparations from the rabbit renal inner medulla and the ram seminal vesicle. Metabolism was measured by the rate of decrease in absorbance at 400 nm. In these soluble and particulate preparations, FANFT metabolism was observed o...
متن کاملA third fatty acid delta9-desaturase from Mortierella alpina with a different substrate specificity to ole1p and ole2p.
A third gene (Delta9-3) encoding a fatty acid Delta9-desaturase was isolated from the oil-producing fungus Mortierella alpina. The predicted protein of 512 aa shared 53% sequence identity with the two fatty acid Delta9-desaturases, ole1p and ole2p, already described in this organism and contained three histidine boxes, four putative transmembrane domains and a C-terminal cytochrome b(5) fusion ...
متن کاملEnzymatic and genetic characterization of firefly luciferase and Drosophila CG6178 as a fatty acyl-CoA synthetase.
Recently we found that firefly luciferase is a bifunctional enzyme, catalyzing not only the luminescence reaction but also long-chain fatty acyl-CoA synthesis. Further, the gene product of CG6178 (CG6178), an ortholog of firefly luciferase in Drosophila melanogaster, was found to be a long-chain fatty acyl-CoA synthetase and dose not function as a luciferase. We investigated the substrate speci...
